What is Sermorelin?

Sermorelin is a synthetic GHRH (1-29) fragment, the shortest sequence that retains full GHRH activity. It is used in pituitary somatotroph culture to study cAMP accumulation and growth hormone release, and NuVion stocks it in Australia as a 5mg lyophilised vial.

Sermorelin structure and research use

Sermorelin is the first 29 amino acids of human growth hormone-releasing hormone, with an amide group at the C-terminus. That is the shortest stretch of the 44 amino acid hormone that keeps full activity at the GHRH receptor. In the laboratory it is used in pituitary somatotroph culture for work on cAMP accumulation and growth hormone release.

What is Sermorelin used for in research?
It is the standard native-sequence agonist for the GHRH receptor. Laboratories use it to define receptor pharmacology in transfected cell lines, to stimulate GH synthesis and release in pituitary cell cultures, and as the unmodified comparator in stability and structure-activity studies of GHRH analogues.
How does Sermorelin work at the cell level?
The GHRH receptor (GHRHR) is a class B1 G protein-coupled receptor expressed almost exclusively on pituitary somatotrophs. Sermorelin binds it with nanomolar affinity and couples it to Gs. Adenylyl cyclase is activated, intracellular cAMP rises, and protein kinase A (PKA) phosphorylates the transcription factor CREB. The Sermorelin research profile covers the mechanism in full.
Is Sermorelin the full GHRH molecule?
No. Sermorelin is residues 1 to 29 of the 44-residue hormone. Truncation studies showed that this stretch carries the full receptor-activating activity, and that the 30 to 44 segment can be removed without loss of potency in pituitary cell assays.
Why is the C-terminus of Sermorelin amidated?
The C-terminal amide on Sermorelin mirrors the amidated C-terminus of the parent hormone and protects the chain from carboxypeptidases. The amide sits in place of the free carboxylate at the end of the 1 to 29 fragment.
Which positions in the Sermorelin sequence are chemically labile?
Four positions are chemically labile. The Tyr1-Ala2 bond is cleaved by dipeptidyl peptidase IV to give GRF(3-29), Asn8 deamidates to Asp or isoAsp, Asp3 can isomerise through a succinimide intermediate, and Met27 oxidises to methionine sulfoxide.
What is the difference between sermorelin and CJC-1295?
Both are based on GHRH (1-29). Sermorelin is the unmodified sequence. CJC-1295 No DAC carries four amino acid substitutions made for stability in solution. Tesamorelin is the full 44 amino acid sequence with a modified N-terminus. NuVion stocks all three.
