What is GHK-Cu?

GHK-Cu, often called copper peptide, is a naturally occurring copper-binding tripeptide (Gly-His-Lys) found in human plasma. It is used in fibroblast and keratinocyte culture to study extracellular matrix protein expression and skin cell biology, and NuVion stocks it in Australia in 50mg and 100mg vials.

  • Copper(II) complex of the tripeptide Gly-His-Lys
  • Used in fibroblast and keratinocyte culture models
  • Applied in matrix protein and gene expression assays
  • Supplied lyophilised, purity by RP-HPLC and identity by mass spectrometry

GHK-Cu structure and research use

GHK-Cu is the copper(II) complex of glycyl-L-histidyl-L-lysine. The bound copper ion gives the lyophilised powder its blue colour. In the laboratory it is used in fibroblast and keratinocyte models for work on collagen and other extracellular matrix proteins and on gene expression. It is also one of the peptides in the GLOW and KLOW blends.

GHK-Cu vial sizes and prices, 50mg and 100mg

NuVion stocks GHK-Cu in 50mg and 100mg vials. Both hold the same lyophilised peptide, and the 100mg vial works out cheaper per milligram.

GHK-Cu research applications

  • Fibroblast and keratinocyte cultures read for collagen, glycosaminoglycan and decorin synthesis and MMP-2, TIMP-1 and TIMP-2 secretion
  • Copper coordination studies by UV-visible spectroscopy, EPR, potentiometric titration and copper exchange experiments with albumin
  • Boyden chamber and transwell chemotaxis assays using monocytes, mast cells or endothelial cells
  • Gene expression profiling by qPCR or RNA sequencing in cultured cells, including NF-kB reporter assays

Storing and handling GHK-Cu

Before reconstitution
Keep the sealed vial cool, dry and out of direct light. General practice for lyophilised peptides.
After reconstitution
Reconstitute with bacteriostatic or sterile water, store at 2 to 8°C and avoid repeated freeze-thaw cycles. No stability-tested shelf life is published for this product.

GHK-Cu questions

Where can I buy GHK-Cu in Australia?
NuVion stocks GHK-Cu in Australia in 50mg and 100mg vials. Choose a size at the top of this page and check out, and the order is posted from Australian stock on a tracked express service. It is supplied as a research material only and is not on the Australian Register of Therapeutic Goods.
How much does GHK-Cu cost in Australia?
It depends on the vial size. Current prices for both sizes are shown on this page in AUD, and the 100mg vial costs less per milligram than the 50mg.
What is GHK-Cu used for in research?
It is used as a defined copper-delivery peptide in fibroblast and keratinocyte culture, where collagen, glycosaminoglycan and metalloproteinase readouts are measured, and in cell-free work on copper coordination and antioxidant chemistry. It also serves as a reference chemoattractant in migration assays and as a stimulus in gene expression profiling.
How does GHK-Cu work at the cell level?
No dedicated cell-surface receptor for GHK has been identified. Its activity in cell models is described through two overlapping routes: delivery of copper into cells, and direct action of the tripeptide on matrix-producing cells. Copper enters cells through the CTR1 transporter after reduction to Cu(I) and is loaded onto cuproenzymes by chaperone proteins. The GHK-Cu research profile covers the mechanism in full.
How does GHK-Cu hold its copper ion?
Cu(II) is held by three nitrogen donors: the glycine alpha-amino group, the deprotonated amide nitrogen of the Gly-His bond and the histidine imidazole nitrogen. This gives a square-planar geometry with one site left open for a water molecule.
Why is GHK-Cu sometimes recorded as a 2:1 peptide to copper complex?
In the solid state the open coordination site on the copper is taken by the carboxylate of a second GHK molecule. Crystallographic records therefore describe a 2:1 species with a mass of 742.3 g/mol, against 340.4 g/mol for the free tripeptide.
Which buffer components break up the GHK-Cu copper complex?
Chelators such as EDTA or citrate strip the copper, and reducing agents such as DTT or ascorbate reduce it to Cu(I). Phosphate at high concentration can precipitate copper, so HEPES or Tris is used for stock solutions.
What does GHK-Cu stand for?
GHK is the one-letter code for the three amino acids in the peptide: glycine, histidine and lysine. Cu is the copper(II) ion the tripeptide binds. Copper peptide is a common name for the same compound.
Why is GHK-Cu blue?
The colour comes from the copper(II) ion held by the tripeptide. The lyophilised powder is blue, and so is the solution once it is made up.
What form does GHK-Cu arrive in?
As a lyophilised (freeze-dried) powder in a sealed vial. Bacteriostatic water is sold separately.
