What is Epithalon?

Epithalon, also spelled Epitalon, is a synthetic tetrapeptide bioregulator (Ala-Glu-Asp-Gly) from the Khavinson short peptide series. It is used in fibroblast and somatic cell culture to study telomerase activity, telomere length and chromosomal stability, and NuVion stocks it in Australia as a 10mg lyophilised vial.

Epithalon structure and research use

Epithalon is four amino acids long: alanine, glutamic acid, aspartic acid and glycine, written AEDG in one-letter code. It is the synthetic counterpart of a peptide fraction first described in pineal gland extracts, and it sits in the same Khavinson series as the tripeptide Pinealon. In the laboratory it is used in fibroblast and somatic cell models for work on telomerase activity, telomere length and chromosomal stability.

  • Telomere length is tracked across passages by qPCR or terminal restriction fragment analysis with population doubling counts

What is Epithalon used for in research?
Epithalon is used in cell culture studies of telomerase expression and activity, telomere length across passages, chromatin condensation state and transcription of specific genes. It is also used in biophysical work on peptide-DNA and peptide-histone binding, and as a test analyte in RP-HPLC method development for short, poorly retained acidic peptides.
How does Epithalon work at the cell level?
No receptor is known and none is proposed. The working hypothesis of the group that developed the peptide is that short, charged peptides cross the plasma and nuclear membranes and bind DNA or chromatin proteins directly, changing the accessibility of particular promoters. Is Epithalon a fragment of a natural protein?
No. Epithalon does not correspond to a stretch of any one characterised protein. Its four residues were chosen as the most abundant amino acids in Epithalamin, a polypeptide fraction from pineal gland tissue, and the peptide is made by solid-phase synthesis.
Why is Epithalon hard to retain on a C18 HPLC column?
Epithalon is very hydrophilic and carries a net charge of about minus 2 at neutral pH, so it is poorly retained on C18 under standard acidic mobile phases. Methods use low starting organic content, an ion-pairing agent or a polar-embedded stationary phase.
What mass does Epithalon show on mass spectrometry?
The molecular weight of Epithalon is 390.35 g/mol. On electrospray in positive mode the singly charged ion appears near m/z 391, and negative mode gives a stronger signal near m/z 389 because the carboxylate groups ionise readily.
Is Epithalon the same as Epitalon?
Yes. Epithalon and Epitalon are two spellings of the same tetrapeptide, Ala-Glu-Asp-Gly.
