What is Thymulin?

Thymulin is a zinc-dependent nonapeptide thymic hormone. It is used in thymocyte and T-lymphocyte culture to study zinc binding and T-cell differentiation, and NuVion stocks it in Australia as a 10mg lyophilised vial.

Thymulin structure and research use

Thymulin is nine amino acids long, pGlu-Ala-Lys-Ser-Gln-Gly-Gly-Ser-Asn, and is produced by the epithelial cells of the thymus. It was first described as serum thymic factor, which is why older papers call it FTS. The peptide is only active when it carries one zinc ion per molecule. In the laboratory it is used in thymocyte and T-lymphocyte culture for work on zinc binding and T-cell differentiation.

  • NF-κB reporter assays, IκB-α Western blotting and cytokine ELISA in lipopolysaccharide-stimulated epithelial cell lines, with and without zinc
  • T cell surface marker induction is scored on lymphoid precursor cells cultured with the zinc complex

What is thymulin used for in research?
Thymulin is used as a tool compound in three areas: T cell lineage marker assays in cultured precursor cells, NF-κB and cytokine signalling studies in epithelial and macrophage-like cell lines, and metal-binding studies where a nine-residue zinc-dependent peptide is a tractable model. It also serves as a reference standard in RP-HPLC and mass spectrometry method development.
How does Thymulin work at the cell level?
No thymulin receptor has been cloned. Saturable, high-affinity binding sites have been described on T lymphoblastoid cell lines, and binding depends on the zinc-bound form, but the protein responsible has not been identified. Why is zinc added to Thymulin in assays?
Thymulin is active only as a complex with one zinc ion. The metal-free peptide is largely unstructured and inactive in bioassays, and synthetic material is normally supplied metal-free, so the active complex is formed in the assay buffer by adding a zinc salt.
What does the pyroglutamate residue do in Thymulin?
The pyroglutamate ring at the N-terminus of Thymulin removes the terminal positive charge and blocks aminopeptidase access. It forms when an N-terminal glutamine or glutamate cyclises, and it puts the observed mass 17 Da below the linear glutamine-initiated sequence.
Where does zinc bind on Thymulin?
NMR work places the zinc-coordinating groups in the C-terminal Ser-Asn region of Thymulin, together with backbone carbonyl oxygens. The sequence has none of the cysteine or histidine ligands of zinc-finger motifs, so the complex is comparatively labile and chelators such as EDTA strip the metal.
Is thymulin the same as thymosin alpha-1?
No. Thymulin is a nine amino acid peptide that needs zinc to be active. Thymosin Alpha-1 is a separate 28 amino acid thymic peptide. NuVion stocks both, along with Thymogen, a two amino acid thymic peptide.
