Thymosin Alpha-1 structure and research use
Thymosin Alpha-1 is the first 28 amino acids of prothymosin alpha, with an acetyl group on its N-terminus. It was first isolated from a thymic extract known as thymosin fraction 5. In the laboratory it is used in T-lymphocyte and dendritic cell models for work on Toll-like receptor signalling, cytokine expression and T-cell maturation.

- NF-κB and interferon-stimulated response element reporter assays in HEK293 lines transfected with TLR2 or TLR9
- Monocyte-derived dendritic cell cultures are read by flow cytometry phenotyping, cytokine qPCR and ELISA on supernatant
- IDO1 induction is measured by Western blot or kynurenine accumulation, with MyD88 or TLR knockdown controls
- Western blotting tracks p65 nuclear translocation, IκBα degradation and p38 or JNK phosphorylation in antigen-presenting cell lines
