What is VIP?

VIP is a 28 amino acid neuropeptide of the glucagon and secretin family, acting at the VPAC1 and VPAC2 receptors. VIP structure and research use

VIP stands for vasoactive intestinal peptide. It is 28 amino acids long and belongs to the glucagon and secretin family, where its closest relative is PACAP. It acts at two G protein-coupled receptors, VPAC1 and VPAC2. In the laboratory it is used in immune cell, gastrointestinal and respiratory epithelial culture models for work on receptor signalling and cytokine response.

What is VIP used for in research?

How does VIP work at the cell level?

Why does the C-terminal amide matter for VIP?
VIP ends in an amidated asparagine. Removing the amide, as happens when the sequence is made without amidation, sharply reduces receptor affinity, so the amide is a specification point for any preparation used in receptor assays.
How is VIP related to PACAP?
PACAP shares 68 per cent sequence identity with VIP and binds VPAC1 and VPAC2 with comparable affinity, while also activating the PAC1 receptor. Separating VPAC from PAC1 responses generally requires selective analogues or receptor knockdown.
What structure does VIP adopt at a membrane?
In aqueous buffer VIP is largely disordered. With membranes or membrane-mimetic solvents present it folds into an amphipathic alpha helix through residues 7 to 28, with polar and non-polar residues on opposite faces.
What receptors does VIP act on?
VPAC1 and VPAC2, both G protein-coupled receptors. It shares them with the related peptide PACAP.
